Product Description 
Mycoplasma, first discovered in 1898, was firstly classified as virus. There are more than 100 known species be-longing to the genus Mycoplasma. They are the smallest free-living, self-replicating organisms and lack a cell wall so that antibiotics such as penicillin, which interfere with murein formation, are not effective. If not treated, mycoplasma contaminations can lead to diminished cell growth and to the loss of cultures.
MycoXpert is a high efficient antibiotic active at low concentration for a broad range of mycoplasma subspecies. It acts by interference with the DNA replication machinery and thus inhibiting mycoplasma growth. MycoXpert is provided as a 50x concentrate.

Procedure
Treatment with MycoXpert includes three or more cycles (passages). It is recommended to check the culture every week for presence of mycoplasma (e.g. ELISA, fluorescence staining or PCR). Please note, that using PCR for detection may lead to false positive results during treatment with MycoXpert since this method cannot distinguish between living and dead mycoplasma. We recommend using PCR as detection method only approx. 2 weeks after treatment with MycoXpert in order to avoid false positive results. Normally mycoplasma will be removed after 2 to 3 weeks.
 
Concentration
MycoXpert is provided as a 50x concentrate.
Working concentration: 1x. 
Therefore dilute the stock solution 1 : 50 (Dilution 1)
Example: Add 10 ml MycoXpert to 500 ml base medium.
If required increase MycoXpert concentrations as follows in different subsequent steps:
Dilution 2: 1 : 40
Dilution 3: 1 : 30
Dilution 4: 1 : 25
 
Instruction for Use
Dilution 1 (1 : 50)
Remove culture medium from culture vessels, wash cells with PBS and detach cells using Trypsin or Accutase.
Count cells and plate in fresh medium (with MycoXpert).
Cultivate cells for 2 to 3 days in the usual way.
Repeat the passaging cycle at least twice.
Check for mycoplasma contamination (Staining method, cultivation or ELISA).
If mycoplasma positive, check methods (positive and negative controls and evaluate).
If required repeat treatment with increased MycoXpert concentrations (Dilution 2, Dilution 3 or Dilution 4).
